Prometheus: the two-component growth hormone pairing, examined

Prometheus is a named combination of CJC-1295 and ipamorelin, the most common pairing in the peptide market. Both act on growth hormone release through different receptors. Human evidence for either is limited, and no controlled trial of the pairing itself was identified.

Overview
This is the simplest grouping in the library, which makes it the clearest example of how component evidence and combination evidence differ. Two substances, two receptors, one shared axis. The pairing is widely marketed on the strength of a mechanistic argument that sounds convincing and has not been tested in an adequately controlled human study of the pair.
Why these are grouped
Releasing hormone analogues and secretagogues act at different receptors on the pituitary. The argument for combining them is that two inputs may produce a more physiologic release pattern than one. That is a reasonable hypothesis. It has not been converted into human outcome evidence for this pairing, and a plausible mechanism is not a result.

A growth hormone releasing hormone analogue studied for a longer signalling window. Human data are limited to small early pharmacology studies.
ipamorelin
A selective secretagogue acting through the ghrelin receptor pathway in preclinical work, with limited human data.
Combination evidence
- No adequately powered controlled human trial of the CJC-1295 and ipamorelin pairing was identified.
- Widespread market use is not evidence. It reflects availability and marketing rather than published results.
- No published safety data describe the pairing over extended periods.
Small human pharmacology studies describe growth hormone response to releasing analogues. Ipamorelin's selectivity comes largely from preclinical work. Neither body of work tells you what the two do together, and neither establishes an outcome such as changed body composition, better sleep, or faster recovery.

- Two receptor pathways converge on pituitary growth hormone release, which is the mechanistic basis for pairing them.
- Preclinical work suggests ipamorelin has a narrower hormonal footprint than earlier secretagogues, which is why it is chosen over them.
What is not established
- Whether the pairing changes body composition, recovery, or sleep in healthy adults.
- Whether it offers anything a single component does not.
- Long-term safety of sustained pituitary stimulation.
- Effects in older adults, who are frequently the people asking about it.

- What would you expect this to change, and how would we measure it?
- Is there evidence that the pairing beats one component alone?
- What laboratory work is appropriate before and during?
- What would you do instead if I want better recovery and sleep?
- At what point would you stop?
